The genetic diversity of 77 barely genotypes were evaluated using seven AFLP combination primers. A total of 245 bands were generated which 227 were polymorph with average of 32.42 polymorphic band per marker. Average percentage of polymorphism and polymorphic information content were 92.37% and 0.43, respectively. General evaluation of the genetic diversity statistics including the number of effective alleles, Nei’s gene diversity and Shannon’s index showed that three primer combinations of E90-M160, E100-M160 and E100-M150 had higher values than the other combinations and had a more obvious role in distinguishing of the studied genotypes. Therefore, these primers are recommended to further studies on barely using AFLP makers. Cluster analysis using the neighbor joining method assigned genotypes to six groups. This type of grouping was nearly consistent with grouping of the genotypes in term of cold, warm or temperate climate, but not as comprehensive accordance. The analysis of population genetic structure using STRUCTURE software showed that population structure is separable into two main sub-populations with 21 and 35 genotypes, respectively, and 21 genotypes were identified as mixed genotypes. Genotypes of each sub-population had most similarity for allele frequencies and genetic structure and are different from the other groups. Results from this research can be used in future breeding programs such as association mapping.}, keywords = {Genetic diversity statistics,Genetic structure,Primer combinations}, title_fa = {بررسی ساختار جمعیت و تنوع ژنتیکی ژنوتیپ‌های جو با استفاده از نشانگرهای مولکولی AFLP}, abstract_fa = {کاهش تنوع ژنتیکی موجب آسیب‌پذیری شدید محصولات زراعی در برابر تنش‌های محیطی، آفات و بیماری‌ها و در نتیجه کاهش عملکرد می‌شود. در این آزمایش تنوع ژنتیکی 77 ژنوتیپ جو با استفاده از هفت جفت آغازگر AFLP بررسی شد. در مجموع 245 نوار ایجاد شد که از بین آنها 227 نوار چندشکل بودند و میانگین تعداد نوارهای چندشکل، 32.42 نوار در هر نشانگر بود. میانگین درصد چندشکلی و محتوای اطلاعات چندشکل نیز به ترتیب 92.37 درصد و 0.43 بود. بررسی کلی آماره‌های تنوع ژنتیکی از جمله تعداد آلل مؤثر، تنوع ژنی نی و شاخص شانون نشان داد که از بین هفت ترکیب آغازگری، سه ترکیب E90-M160 ، E100-M160 و E100-M150 نسبت به سایر ترکیب­ها مقادیر بالاتری را به خود اختصاص دادند و در حقیقت در تمایز ژنوتیپ‌ها نقش بارزتری ایفا نمودند و از این­رو، این ترکیب­ها برای مطالعات بعدی روی جو با استفاده از نشانگرهای AFLP پیشنهاد می‌شوند. تجزیه خوشه‌ای با استفاده از روش اتصال همسایه­ها، ژنوتیپ‌ها را به شش گروه تقسیم کرد که با تقسیم‌بندی ژنوتیپ‌ها از لحاظ تیپ سرد، گرم یا معتدله مطابقت نسبی داشت، اما این مطابقت جامع نبود. از سوی دیگر، تجزیه ساختار ژنتیکی ژنوتیپ­ها با استفاده از نرم‌افزار STRUCTURE نشان داد که ساختار جمعیت به دو زیرجمعیت اصلی قابل تفکیک است. بر این اساس 21 و 35 ژنوتیپ به‌ترتیب به گروه 1 و 2 منتسب و 21 ژنوتیپ نیز به عنوان مخلوط شناخته شدند. ژنوتیپ‌های مربوط به هر زیر جمعیت از لحاظ ساختار و فراوانی آللی بیشترین شباهت را با هم داشتند و متفاوت از سایر گروه‌ها بودند. اطلاعات حاصل از پژوهش حاضر را می‌توان در برنامه‌های اصلاحی بعدی همانند مکان‌یابی ارتباطی استفاده کرد.}, keywords_fa = {آماره‌های تنوع ژنتیکی,ترکیبات آغازگری,ساختار ژنتیکی}, url = {https://cr.guilan.ac.ir/article_1434.html}, eprint = {https://cr.guilan.ac.ir/article_1434_8699644f82ef29659b893f4443895d90.pdf} }
