بررسی برخی ویژگی های جوانه زنی، صفات بیوشیمیایی و میزان بیان ژنهای آنتیاکسیدان در ژنوتیپ های گندم نان در مرحله گیاهچه ای تحت تنش فلز جیوه
آﻟﻮدﮔﯽ ﺑﺎ ﻓﻠﺰات ﺳﻨﮕﯿﻦ یکی از ﻣﻬمترین ﻋﻮاﻣﻞ ﺟﻬﺎﻧﯽ آﻟﻮدﮔﯽ ﺧﺎک است. آﻻﯾﻨﺪهﻫﺎی ﻓﻠﺰی ﺑﻪدلیل ﻏﯿﺮ ﻗﺎﺑﻞ ﺗﺠﺰﯾﻪ ﺑﻮدن و آثار ﻓﯿﺰﯾﻮﻟﻮژیک آنها ﺑﺮ ﻣﻮﺟﻮدات زﻧﺪه و اﻧﺴﺎن، ﺣﺘﯽ در ﻏﻠﻈﺖﻫﺎی کم سرطانزا هستند. ﺟﻮاﻧﻪزﻧﻲ ﺑﺬر، ﻳﻜﻲ از ﻣﻬﻢﺗﺮﻳﻦ ﻣﺮاﺣﻞ در ﭼﺮﺧﻪ زﻧﺪﮔﻲ ﮔﻴﺎهان است ﻛﻪ ﺑﻪ عوامل ﻣﺤﻴطی از ﻗﺒﻴﻞ تنشﻫﺎی زﻳﺴﺘﻲ و ﻏﻴﺮزﻳﺴﺘﻲ واﻛﻨﺶ ﻧﺸﺎن میدﻫﺪ. هدف از اجرای این آزمایش، ارزیابی ویژگیهای جوانهزنی، صفات بیوشیمیایی و بیان ژنهای آنتیاکسیدان در ژنوتیپهای گندم نان در مرحله گیاهچهای تحت تنش فلز جیوه بود. آزمایش بهصورت کرتهای خرد شده در قالب طرح پایه کاملاً تصادفی با سه تکرار اجرا شد که در آن تنش جیوه در چهار سطح (شامل بدون کاربرد بهعنوان شاهد و کاربرد 5، 10 و 15 میکرومولار کلرید جیوه) و ژنوتیپ گندم در سه سطح (شامل گنبد، مروارید و لاین N9108) بهترتیب بهعنوان عامل اصلی و فرعی آزمایش در نظر گرفته شدند. نتایج نشان داد که با افزایش میزان تنش کلرید جیوه، شاخصهای جوانهزنی و صفات مورفولوژی و بیوشیمیایی در هر سه ژنوتیپ گندم روندی کاهشی داشت و میزان این کاهش بهطور معنیداری در ژنوتیپهای مختلف، متفاوت بود. لاین N9108 از لحاظ همه صفات اندازهگیری شده، شرایط بهتری نسبت به دو ژنوتیپ دیگر تحت تنش فلز جیوه داشت. میزان بیان ژن سوپراکسید دیسموتاز و پراکسیداز نیز در لاین N9108 بیشتر از ژنوتیپهای دیگر بود که میتواند نشاندهنده برتری این لاین نسبت به دو ژنوتیپ دیگر گندم تحت شرایط تنش کلرید جیوه باشد.

شناسایی بیوانفورماتیکی و بررسی بیان miRNA های دخیل در نمو دانه ارقام گندم ایرانی با کیفیت نانوایی متفاوت
کیفیت نانوایی گندم (Triticum aestivum L.) بهشدت وابسته به حضور و ترکیب پروتئینهای گلوتن در دانه است. در این مطالعه، کیفیت نانوایی چهار رقم گندم ایرانی (مروارید، سرداری، پارسی و سپاهان) بر اساس آنالیزهای فارینوگرافی و شیمیایی آرد و حجم ویژه نان بهترتیب از بیشترین به کمترین کیفیت رتبهبندی شد. بهمنظور شناسایی و مقایسه miRNA های دخیل در نمو دانه بین گندمهای ایرانی با کیفیت نانوایی خوب و ضعیف، دادههای RNA-seq هفت رقم گندم (چهار رقم با کیفیت خوب و سه رقم با کیفیت ضعیف) در دو مرحله نموی دانه از پایگاه اطلاعاتی NCBI پیادهسازی و آنالیز شدند. از miRNA های شناسایی شده، پنج miRNA حفاظتشده انتخاب و الگوی بیان آنها با استفاده از روش Real Time PCR در دو مرحله نموی دانه (10 و 20 روز پس از گردهافشانی) در چهار رقم گندم ایرانی بررسی شد. نتایج نشان داد که الگوی بیان miR159a، miR164a، miR172a و miR827a طی نمو دانه در همه ارقام مورد بررسی روند افزایشی داشت، در حالیکه الگوی بیان miR396c روندی کاهشی نشان داد. miR164a و miR396c بیشترین و کمترین بیان نسبی را بهترتیب در ارقام سپاهان و سرداری (بهترتیب با کمترین و بیشترین وزن هزار دانه) نشان دادند که میتواند در نتیجه نقش تنظیمکنندگی منفی آنها بر اندازه دانه باشد. همچنین، با افزایش کیفیت نانوایی ارقام مورد بررسی، بیان نسبی miR172a و miR159a که بهترتیب در سنتز نشاسته و پروتئینهای ذخیرهای دانه دخیل هستند، کاهش و بیان miR827a که در فرایند انتقال مجدد نیتروژن از برگ به دانه دخیل است، افزایش یافت که نشاندهنده آثار منفی miR172a و miR159a و مثبت miR827a بر کیفیت نانوایی ارقام گندم میباشد.

پایداری عملکرد دانه ژنوتیپهای گندم دوروم در مناطق نیمهگرمسیر دیم
آگاهی از برهمکنش ژنوتیپ × محیط به بهنژادگران کمک میکند تا بتوانند بهترین ژنوتیپها را برای مناطق مختلف انتخاب کنند. هدف از این پژوهش، دستیابی به ژنوتیپهای پرمحصول گندم دوروم از بین چند لاین برگزیده بود تا بتوانند سازگار با شرایط آب و هوایی مناطق دیم گرمسیری و نیمهگرمسیری و برتر از ارقام شاهد منطقه باشند. در این پژوهش، 16 ژنوتیپ گندم دوروم برگزیده از آزمایشهای پیشرفته مقایسه عملکرد، بههمراه دو رقم شاهد دهدشت و سیمره، در قالب طرح بلوکهای کامل تصادفی با چهار تکرار بهمدت سه سال زراعی (95-1392) در چهار منطقه (گچساران، گنبد، خرمآباد و مغان) مورد مطالعه قرار گرفتند. تجزیه واریانس مرکب دادهها نشان داد که آثار اصلی ژنوتیپ، سال و مکان و برهمکنش سال × مکان و ژنوتیپ × سال × مکان در سطح احتمال یک درصد معنیدار بود. تجزیه واریانس AMMI نیز نشان داد که چهار مولفه اصلی اول معنیدار بودند و در مجموع حدود 81 درصد از تغییرات برهمکنش ژنوتیپ × محیط را توجیه کردند. بیشترین عملکرد دانه در ژنوتیپ شماره 6 (3592 کیلوگرم در هکتار) و کمترین میزان آن در ژنوتیپ شماره 18 (3229 کیلوگرم درهکتار) مشاهده شد. محیطهای 10، 8، 11 و 12 با بردارهای بلند، از قدرت تفکیک بالایی برخوردار بودند. نتایج این تحقیق بر اساس شاخصهای AMMI نشان داد که ژنوتیپهای 3، 5، 6، 13 و 15 با میانگین عملکرد دانه بالاتر از میانگین کل در بیشتر محیطها، بهعنوان ژنوتیپهای پایدار و برتر آزمایش بودند. از بین آنها، ژنوتیپ شماره 13 با توجه به دارا بودن سازگاری عمومی مناسب، میتواند بهعنوان ژنوتیپ امیدبخش و نامزد معرفی رقم جدید برای مناطق دیم گرمسیر و نیمهگرمسیر باشد.

ارزیابی ویژگی های مرفوفنولوژیک گندم های هگزاپلوئید مصنوعی
گندمهای هگزاپلوئید مصنوعی، آمفیپلوئید حاصل از تلاقی بین گندم دوروم و گونههای آجیلوپس، منابع ارزشمندی برای افزایش تنوع ژنتیکی گندم و استفاده از آللهای بیرون از خزانه ژنی اولیه گندم هستند. در این تحقیق، تعداد 50 لاین گندم هگزاپلوئید مصنوعی همراه با پنج رقم گندم نان (آنفارم4، ارگ، افق، نارین و بککراس روشن) بهعنوان ارقام شاهد آزمایش برای صفات مورفوفنولوژیک مختلف در آزمایشگاه و مزرعه تحقیقاتی دانشکده کشاورزی دانشگاه بیرجند مورد ارزیابی قرار گرفتند. نتایج تجزیه واریانس دادهها نشان داد که اختلاف آماری معنیداری بین ژنوتیپهای مختلف برای همه صفات مورد مطالعه بهجز تعداد پنجه وجود داشت که نشاندهنده وجود تنوع ژنتیکی بین ژنوتیپها است. وراثتپذیری برای صفات مختلف در محدوده 26 تا 90 درصد برآورد شد. مقایسه میانگین بین لاینهای گندم مصنوعی و ارقام شاهد نشان داد که برای بیشتر صفات مطالعهشده بهجز طول سنبله، تعداد پنجه و وزن هزار دانه، اختلاف آماری معنیداری وجود داشت. تجزیه خوشهای بر اساس روش حداقل واریانس وارد، ژنوتیپها را به سه گروه دستهبندی کرد. نمودار بایپلات بر اساس دو مولفه اصلی که 09/53 درصد از تغییرات کل دادهها را توجیه کردند نیز گروهبندی حاصل از تجزیه خوشهای را تایید کرد. نتایج این تحقیق نشان داد که اگرچه برتری کلی با ارقام شاهد بود، اما تنوع ژنتیکی زیادی برای بسیاری از صفات مورد مطالعه در لاینهای گندم مصنوعی مشاهده شد و بنابراین از لاینهای دارای صفات برتر میتوان بهعنوان والدین انتخابی برای تولید ارقام گندم جدید در برنامههای بهنژادی آینده استفاده کرد.

تجزیه همبستگی کانونیک صفات فیزیولوژیک با صفات فنولوژیک و ریشهای در ژنوتیپهای مختلف گندم نان
بهمنظور برآورد همبستگی و روابط بین صفات مختلف در گندم، 30 ژنوتیپ گندم نان در دو آزمایش جداگانه در گلخانه و مزرعه تحت دو شرایط بدون تنش و تنش خشکی در مرحله شروع گلدهی مورد مطالعه قرار گرفتند. طرح آزمایش در هر دو شرایط گلخانه و مزرعه، طرح کرتهای خردشده بود که در آن تنش خشکی بهعنوان فاکتور اصلی و ژنوتیپهای گندم بهعنوان فاکتور فرعی آزمایش در نظر گرفته شدند، اما طرح پایه در گلخانه طرح کاملاً تصادفی و در مزرعه طرح بلوکهای کامل تصادفی هر دو با سه تکرار بود. نتایج حاصل از تجزیه همبستگی کانونیک نشان داد که همبستگی معنیداری در هر دو سطح بدون تنش و تنش خشکی بین جفت متغیرهای کانونیک حاصل از صفات فیزیولوژیک با صفات ریشهای و فنولوژیک وجود داشت، بهطوریکه در شرایط مزرعه، تابع کانونیک اول برای صفات فیزیولوژیک (V1) بیشتر متأثر از تعداد روز تا 50 درصد گلدهی و سرعت پر شدن دانه بود، ولی برای صفات فنولوژیک (W1) بیشتر تحت تأثیر صفات کاتالاز، پراکسیداز، مالون دیآلدهید، میزان پراکسید هیدروژن برگ و محتوای پرولین قرار داشت. در مقابل در شرایط گلخانه، تابع کانونیک اول برای صفات ریشهای (V1) بیشتر تحت تأثیر میزان پرولین، میزان H2O2 برگ، MDA، پراکسیداز، کاتالاز، غلظت کلروفیل کل و کلروفیل a قرار گرفت، در حالیکه برای صفات فیزیولوژیک (W1) بیشتر متأثر از صفت وزن خشک ریشه بود. نتایج این آزمایش نشان داد که بهمنظور گزینش ارقام متحمل به خشکی تحت هر دو شرایط بدون تنش و تنش خشکی، افزایش وزن خشک ریشه از طریق افزایش غلظت MDA، میزان H2O2 برگ، میزان پرولین، پراکسیداز و کاتالاز و کاهش کلروفیل کل و کلروفیل a تحت شرایط گلخانه و افزایش سرعت پر شدن دانه از طریق افزایش کاتالاز، پراکسیداز و پرولین و کاهش غلظت MDA و H2O2 برگ تحت شرایط مزرعه میتوانند بهعنوان معیارهای گزینشی مناسب در نظر گرفته شوند.

بررسی کارایی علفکش کندرهاشونده پیرازوسولفوروناتیل + پرتیلاکلر در کنترل علفهای هرز برنج در کشت نشایی
تنوع علفهای هرز شالیزار، موجب استفاده از مخلوط علفکشهای با مکانیزم عمل متفاوت جهت افزایش کارایی مدیریت شیمیایی شده است. علفکش جدید پیرازکلر (Pirazchlor, TB 17%) حاصل اختلاط پیرازوسولفوروناتیل از سولفونیلاورهها با پرتیلاکلر از کلرواستامیدها است که بهصورت قرصهای حدود پنج گرمی کندرهاشونده برای کنترل علفهای هرز برنج در کشت نشایی معرفی شده است. در این پژوهش، کارایی مقادیر 191، 383، 574 و 765 گرم ماده موثره در هکتار (g ai.ha-1) پیرازکلر در مقایسه با علفکشهای رایج پرتیلاکلر (EC 50%, 750 g ai.ha-1)+ بنسولفورونمتیل (بنسولفورون، DF 60%, 35 g ai.ha-1)، تیوبنکارب (EC 50%, 2500 g ai.ha-1)+ بنسولفورون، تریافامون+ اتوکسیسولفورون (کانسیل، WG 30%, 30 g ai.ha-1) بههمراه دو تیمار شاهد (بدون کنترل و وجیندستی) بر کنترل علفهای هرز و عملکرد برنج رقم هاشمی مورد بررسی قرار گرفت. آزمایش در قالب طرح بلوکهای کامل تصادفی با سه تکرار در مزارع پژوهشی موسسه تحقیقات برنج کشور در گیلان و مازندران در سال 1397 انجام شد. نتایج نشان داد که کنترل علفهای هرز و عملکرد برنج تحت تأثیر تیمار، مکان و برهمکنش تیمار در مکان قرار گرفت. در مازندران، زیستتوده و تراکم علفهای هرز در حدود یکدهم گیلان و کارایی تمام علفکشها مساوی یا بیشتر از 95 درصد بود. در گیلان، کارایی علفکشها در کاهش تراکم سوروف (که بیشتر یا مساوی 88 درصد بود)، بیش از پیزور (با 51 تا 97 درصد) بود. کارایی پیرازکلر در دزهای 383 گرم و بیش از آن، مشابه یا بهتر از علفکشهای رایج بود. خسارت اقتصادی علف هرز در تیمار شاهد بدون کنترل در مازندران و گیلان بهترتیب 25 و 78 درصد نسبت به تیمار شاهد وجیندستی برآورد شد. بیشترین عملکرد شلتوک در گیلان 4450 و مازندران 5001 کیلوگرم در هکتار بود که بهترتیب از دزهای 765 و 363 گرم پیرازکلر بهدست آمد و مشابه یا بیشتر از تیمارهای علفکش رایج و در حدود 10 درصد بیشتر از تیمار شاهد وجین دستی بود.

ارزیابی اثربخشی طرح تجهیز و نوسازی اراضی شالیکاری بر توسعه مکانیزاسیون زراعت برنج در استان گیلان (مطالعه موردی شهرستان املش)
تجهیز و نوسازی اراضی شالیکاری از اقدامات زیربنایی مهم در جهت تدارک زیرساختهای زراعت مکانیزه برنج است. در این راستا، بهمنظور ارزیابی اثربخشی طرح تجهیز و نوسازی اراضی شالیکاری بر توسعه کشت مکانیزه برنج، پژوهشی به روش پیمایشی در شهرستان املش در شرق استان گیلان انجام شد. بر این اساس، حجم نمونه با استفاده از جدول حداقل حجم نمونه بارتلت برای دو گروه از شالیکاران دارای اراضی شالیکاری تجهیزنشده و تجهیزشده، بهترتیب 105 و 160 نفر تعیین و نمونههای تحقیق با استفاده از روش نمونهگیری طبقهای چندمرحلهای انتخاب شدند. پرسشنامه محققساخته بهعنوان ابزار گردآوری دادهها طراحی و مورد استفاده قرار گرفت. نتایج تحقیق نشان داد که درجه مکانیزاسیون عملیات کاشت و برداشت برنج در اراضی تجهیزشده در مقایسه با اراضی تجهیزنشده، در هر سه گروه اراضی کوچک (کمتر از نیم هکتار)، متوسط (نیم تا یک هکتار) و بزرگ (بیش از یک هکتار) بیشتر بود. تنها درجه مکانیزاسیون عملیات وجین مکانیکی در شالیزارهای بزرگ تجهیزشده 29/10 درصد و در سایر موارد صفر بود. در میان عملیات زراعی در اراضی تجهیزشده، بالاترین درجه مکانیزاسیون بعد از خاکورزی (100 درصد) مربوط به برداشت برنج (43/56، 53/73 و 62/72 درصد بهترتیب برای شالیزارهای کوچک، متوسط و بزرگ) بود. بیشترین سهم ظرفیت مکانیزاسیون کل در اراضی تجهیزنشده را عملیات خاکورزی و در اراضی تجهیزشده، عملیات برداشت بهخود اختصاص دادند. بنابراین، صرفنظر از پایینبودن درجه مکانیزاسیون عملیات وجین مکانیکی، درجه مکانیزاسیون سایر عملیات زراعی کشت برنج در اراضی تجهیزشده در منطقه مورد مطالعه در وضعیت مناسبی قرار داشت. از اینرو، ضروری است سازمان جهاد کشاورزی استان گیلان با بکاربستن راهکارهای کاربردی، درجه مکانیزاسیون عملیات وجین و نشای مکانیکی برنج در منطقه املش گیلان را نیز ارتقا دهد. همچنین، بهمنظور بهبود ظرفیت مکانیزاسیون در اراضی شالیکاری تجهیزشده، پیشنهاد میشود کارشناسی دقیق اندازه ماشینهای کشاورزی مورد نیاز بهویژه ماشینهای برداشت برنج مورد توجه ویژه قرار گیرد تا به این ترتیب، ظرفیت مکانیزاسیون تا حد ممکن کاهش یابد.
