تاثیر تنش کم آبی بر صفات زراعیو فعالیت آنزیم سوپراکسید دیسموتازدرسه هیبرید ذرت (Zea mays L.)

چکیده

به­منظور بررسی اثر تنش کم­آبی بر سه هیبرید ذرت (SC704، SC740 و NS640)، آزمایشی به­صورت کرت­های خرد شده در قالب بلوک­های کامل تصادفی با چهار تکرار تحت شرایط بدون تنش و تنش کم­آبی (قطع آبیاری قبل از گلدهی) در ایستگاه تحقیقات کشاورزی دانشگاه تبریز در سال زراعی 92-1391اجرا شد. تجزیه الکتروفورزی برای فعالیت سوپراکسید دیسموتاز (Mn-SOD) با استفاده از ژل­های پلی­آکریلامید هشت درصد انجام گرفت و میزان "مساحت × شدت" نوارهای روی ژل با استفاده از نرم­افزار MCID ارزیابی و به­عنوان فعالیت آنزیمی ثبت شد. میزان مالون دی­آلدئید (MDA) نیز تحت دو شرایط آزمایشی در هیبریدهای مورد مطالعه به­عنوان شاخص اکسیداسیون سلولی ارزیابی شد. نتایج نشان داد که تنش کم­آبی به­طور معنی­داری سبب کاهش صفات عملکرد دانه، وزن صد دانه، طول بلال، قطر بلال، ارتفاع بوته، قطر بوته، دمای برگ، شاخص کلروفیل، تعداد شاخه کاکل و طول کاکلدر هیبریدهای مورد مطالعه شد. با افزایش تنش­ کم­آبی، شاخص اکسیداسیون سلولی (MDA) در سینگل­کراس 704 افزایش نشان داد. میزان فعالیت دنسیومتریک Mn-SOD نیز در سینگل­کراس 704 تحت تنش کم­آبی افزایش یافت که نشان­دهنده افزایش رادیکال­های آزاد اکسیژن تحت تنش کم­آبی بود. نتایج حاصل از این تحقیق نشان داد که سینگل­کراس 704متحمل به تنش کم­آبی است.

To study the effects of water deficit stress on three maize hybrids (SC704, SC740 and NS640), an experiment was conducted in split plot based on randomized complete block design with four replications under non-stress and water deficit stress conditions at the Agricultural Research Station of University of Tabriz, Tabriz, Iran, in 2013. Electrophoretic analysis was performed to evaluate the enzymatic activity of superoxide dismutase (Mn-SOD) by using 8% slab polyacrylamide gels and “density × area” values of the electrophoretic bands onto gels were scored asenzymatic activity using MCID software. Malondialdehyde (MDA) content was also evaluated as cellular oxidation index in the studied hybrids under normal and water deficit stress. The results indicated that water deficit stress significantly decreased grain yield, 100-grain weight, ear diameter, ear length, plant height, plant diameter, leaf temperature, chlorophyll index, tassel branch number and tassel length. The cellular oxidation level (MDA) increased in SC704 by increasing water deficit stress. TheMn-SOD activity also increased in SC704 under water deficit showing an increase inoxygen free radicals. The results of this study showed that SC704 was tolerant to water deficit stress.
